THE SIMULTANEOUS OCCURRENCE OF THE BCR-ABL TRANSLOCATION AND THE Jak2V617F MUTATION. DETECTION AND DYNAMICS EVOLUTION IN A CASE OF CHRONIC MYELOPROLIFERATIVE NEOPLASM

چکیده
Although the discovery of the BCR-ABL transcript and of the Jak2 V617F mutation lead to important clarifications, the pathogenesis of chronic myeloproliferative neoplasms remains extremely complex and still presents several unknowns. Our study reports the case of a 52 years old man with chronic myeloid leukemia (BCR-ABL positive) who, after 4 years of targeted therapy with tyrosine kinase inhibitors, also developed PV (Jak2 V617F positive). As such, our findings negate the theory according to which the two genetic transformations and the two different pathologies defined by them, respectively, were mutually exclusive.

Mechanical Trapping of DNA in a Double-Nanopore System.
Nanopores have become ubiquitous components of systems for single-molecule manipulation and detection, in particular DNA sequencing where electric field driven translocation of DNA through a nanopore is used to read out the DNA molecule. Here, we present a double-pore system where two nanopores are drilled in parallel through the same solid-state membrane, which offers new opportunities for DNA...
متن کامل3D structural integrity and interactions of single-stranded protein-binding DNA in a functionalized nanopore.
Biomarker-binding nucleotide sequences, like aptamers, have gained recent attention in cancer cell isolation and detection works. Self-assembly and 3D conformation of aptamers enable them to selectively capture and bind diseased cells and related biomarkers. One mode of utilizing such an extraordinary selective property of the aptamers is by grafting these in nanopores. Coating the inside walls...
متن کاملThe Secretion of IL-22 from Mucosal NKp44+ NK Cells Is Associated with Microbial Translocation and Virus Infection in SIV/SHIV-Infected Chinese Macaques
Microbial translocation (MT) causes systemic immune activation in chronic human immunodeficiency virus (HIV) infection. The role of a novel subtype of innate lymphoid cells, the NKp44(+) NK cells, in HIV/simian immunodeficiency virus- (SIV-) induced MT remains unknown. In this study, 12 simian-human immunodeficiency virus- (SHIV-) infected macaques were chosen and split into two groups based on...
متن کاملDynamics of DNA translocation through an attractive nanopore.
We investigate the dynamics of single-stranded DNA translocation through a nanopore driven by an external force using Langevin dynamics simulations in two dimensions to study how the translocation dynamics depend on the details of the DNA sequences. We consider a coarse-grained model of DNA built from two bases A and C, having different base-pore interactions, e.g., a strong (weak) attractive f...
متن کاملIntrinsic Stepwise Translocation of Stretched ssDNA in Graphene Nanopores
We investigate by means of molecular dynamics simulations stretch-induced stepwise translocation of single-stranded DNA (ssDNA) through graphene nanopores. The intrinsic stepwise DNA motion, found to be largely independent of size and shape of the graphene nanopore, is brought about through alternating conformational changes between spontaneous adhesion of DNA bases to the rim of the graphene n...
